How to edit .img file?

It's not iso, but img. How can img edit the content?

If you are already running an EASY, merely open the filemanager to the folder with the image, and double-click the image file.

Ce bash script de Jafadmin permet de monter/démonter une img ,très utile.

How to open and edit in Puppy? How to package IMG after editing? Is there any software to edit IMG?

You are not providing enough information. Are you running EasyOS when wanting to edit the .img file, as Flash has asked you?

You have posted in the EasyOS section, so we assume answer is yes. So why have you posted "How to open and edit in Puppy" -- in that case, you need to ask elsewhere in the forum.

In EasyOS, there is a script that will open up a .img file, and offer to view it read-only or editable. This script can be run by clicking on the .img file, or, if you open a terminal where the .img file is located:
